The reason for these differences is mainly because of how MPTP is changed into its more dangerous form, MPP+, inside the body. This process varies between species.

In rats, an enzyme called MAO-B helps convert MPTP to MPP+ but also acts as a protective barrier in the brain. In primates, including humans, this enzyme is found mostly in certain brain cells called astrocytes, where it helps turn MPTP into the harmful MPP+. MPP+ then gathers in the brain cells that manage movement, which can lead to their damage.

Scientists are still figuring out exactly how MPP+ causes this damage. It might turn into a harmful free radical, interfere with energy production in cells or cause harmful levels of calcium to build up, leading to cell death.
